The Alpha‑Tec Hypo-Osmotic Swelling (HOS) test is designed to evaluate the functional integrity of sperm plasma membranes. This integrity is crucial for various sperm functions, including motility, capacitation, acrosome reaction, and binding to the Zona Pellucida, all of which can influence fertilizing capacity. The HOS test operates on the premise that in hypo-osmotic conditions, normal sperm tails experience fluid influx, leading to coiling and swelling. Conversely, dead spermatozoa undergo excessive swelling to the point of membrane rupture, causing their tails to straighten. A key advantage of the HOS test is its ability to identify live sperm without terminating them, enabling their use in therapeutic methods like Intra Cytoplasmic Sperm Injection (ICSI).
